Article Snippet: To remove any 5′ m 7 G caps, 500 ng of the SAP-treated RNA was treated with 12.5 units RNA 5’ Pyrophosphohydrolase (RppH; New England BioLabs; Ipswitch, MA, United States) in 1X T4 RNA Ligase Buffer (New England BioLabs; Ipswitch, MA, United States) supplemented with 20 units RNaseOUT (Invitrogen; Carlsbad, CA, United States) in a total reaction volume of 10 μL for 1 h at 37°C and stored at –20°C overnight.
